Service Overview

Eton uses Oxford Nanopore Technology (ONT) MinION devices, flow cells, and library preparation kits to perform whole-plasmid sequencing. Data is assembled on the EPI2ME platform using the wf-clone validation and wf-amplicon workflows.

Use Cases

  • What technology should I use for my sequencing project?
    • Use Nanopore for:
      • Sequencing whole plasmids to validate important backbone sequences, especially for large scale plasmid preps.
      • Sequencing large inserts without having to “primer walk”.
      • Sequencing ITRs and other complex features.
    • Use Sanger for:
      • Sequencing or screening large number of clones, plasmids, or PCR fragments using one or two primers to cover a small region of interest and identify “hits”.
      • Sequencing directly from colonies on agar (or glycerol stocks), especially where the validated clone is planned for scale-up by Eton’s DNA plasmid prep service
      • Sequencing templates containing poly-A or other homopolymeric regions.

Sequencing Service Types

  • Purified Plasmid DNA.
  • Purified PCR fragments from 500 bp to 5 kb.

Sample Prep Guidelines

For plasmids 25 kb and less:

  • Required submission format: PCR strip tubes.
  • Required concentration: 40 ng/u.
  • Required minimum volume: 6 uL.
  • Purified sample with no primers or dye added.

For PCR amplicons between 500 bp and 5 kb:

  • Required submission format: PCR strip tubes.
  • Required concentration: 40 ng/u.
  • Required minimum volume: 6 uL.
  • Purified sample with no primers or dye added.
  • At this time, we cannot accept mixtures or pools of PCR amplicons of varying sequences.

Data Delivered

  • fasta of the consensus sequence.
  • fastq of the consensus sequence.
  • genbank (gbk) plasmid map with annotations.
  • QC report showing:
    • Assembled sequence lengths and mean quality score.
    • Read statistics, including read-length histograms.

How to Order

  1. If you don’t have an account, please Register.
  2. Select “Place Order” under “Nanopore Sequencing”.
  3. On the “Order Information” page, select how you’d like to submit your samples and click “Next Step”.
  4. On the “Results Delivery” page, select how you’d like to receive your data and click “Next Step”.
  5. On the “Order Details” page, enter your sample information: Sample Name, Sample Concentration (ng/uL), and Sample Size (kb) and click on “Next Step”
    1. Please refer to Sample Prep Guidelines for the volumes and concentrations of DNA required.
  6. Review your Order Summary and click on “Next Step”.
  7. On the “Payment Information” page, enter your payment information. Eton accepts a validated PO number or credit card.
  8. Click “Submit Order”.
